Research Overview

Acetyl Hexapeptide-38 (marketed as Adifyline®) is a synthetic hexapeptide designed to address volume loss and deep structural wrinkles associated with subcutaneous fat atrophy in aged skin. Unlike surface-acting anti-wrinkle peptides, Acetyl Hexapeptide-38 targets dermal fibroblasts and pre-adipocytes in the hypodermis, stimulating both collagen synthesis in the dermis and differentiation of pre-adipocytes into mature subcutaneous adipocytes. This dual anabolic action simultaneously rebuilds structural collagen scaffold and restores the cushioning subcutaneous fat layer that thins with chronological aging and photodamage. The N-terminal acetylation of the hexapeptide (Ac-Ser-Val-Val-Val-Arg-Thr-NH2) enhances proteolytic stability and cell membrane permeability relative to the free peptide. In vitro studies demonstrate that Acetyl Hexapeptide-38 significantly upregulates adipogenic markers including PPAR-γ, C/EBPα, and adiponectin in pre-adipocyte cultures, confirming activation of the adipogenic differentiation program.
