Palmitoyl Tripeptide-5 (marketed as SYN-COLL by DSM Nutritional Products) is a lipopeptide consisting of a Lys-Val-Lys tripeptide N-terminally conjugated to a C16 palmitic acid chain. The peptide sequence mimics the thrombospondin-1 (TSP-1) protein's KTTKS collagen-stimulating domain, and it activates the transforming growth factor-β1 (TGF-β1) signaling pathway in dermal fibroblasts by binding to and activating the TSP-1 receptor complex on the fibroblast surface. This triggers downstream SMAD2/3 phosphorylation and transcriptional activation of collagen Type I and III synthesis genes. The palmitic acid moiety serves dual functions: it dramatically enhances transdermal penetration by increasing lipophilicity and enabling partitioning into the lipid bilayer of the stratum corneum, and it stabilizes the peptide against protease degradation, extending its effective residence time in the skin.
