Research Overview

Tripeptide-29 (Gly-Pro-Hyp, where Hyp = hydroxyproline) is the fundamental repeating tripeptide motif that constitutes the structural backbone of all fibrillar collagens. In native collagen molecules, three polypeptide chains each containing repeating Gly-X-Y sequences (where X is frequently proline and Y is frequently hydroxyproline) wrap into the characteristic triple-helical structure. Tripeptide-29 therefore represents both the minimal collagen fragment and a potent biological signal for fibroblast recognition and ECM remodeling. With a molecular weight of only 285 Da, Tripeptide-29 penetrates the stratum corneum and reaches viable dermal layers far more efficiently than high-molecular-weight collagen proteins, which cannot cross the skin barrier. Upon reaching dermal fibroblasts, it activates collagen receptors (including discoidin domain receptors and integrins) and stimulates de novo synthesis of collagen Types I, III, and IV. Simultaneously, it modulates matrix metalloproteinase (MMP) activity, reducing collagenolytic degradation of newly synthesized ECM. The hydroxyproline residue is particularly important for fibroblast recognition, as it signals the presence of mature collagen and triggers anabolic repair responses. Research applications include wound healing (stimulating granulation tissue formation and epithelialization), anti-aging skincare (increasing dermal collagen density and skin firmness), post-procedure recovery (laser, chemical peel, microneedling), and bone/cartilage tissue engineering.
